Rowena tries to take Dean out with some big bad spell, but Dean just reacts as if she had shot a nerf dart at him. Obviously the mark of Cain provides some big time magic protection. So after that failure, she beats herself up and runs to Crowley crying "Dean did it!", trying to get him to kill Dean. After talking to Dean, Crowley realizes that his mother doesn't care a lick for him, anymore than she did when he was a mortal child. So he finally comes to his senses and sends her packing. No doubt she'll put her son on her shlt list right next to the Winchesters now.
